- ItemValidación del proceso de extracción en fase sólida y confirmación simultánea por cromatografía líquida de alta eficiencia de arreglo de diodos de: fenitoína, fenobarbital y carbamazepina en muestras de sangre.(UNIVERSIDAD NACIONAL DE COLOMBIA, 1999) MARTHA PIEDAD MEDINA OSPINA CESAR AUGUSTO PAREDES ORTIZ; Medina Ospina, Martha Piedad; Paredes Ortíz, Cesar Augusto; Ospina de Nigrinis, Stella; Pinzón Peralta, AidéeWe established a precise and accurate high‑performance liquid chromatographic method for the simultaneous assay of phenytoin, phenobarbital and carbamazepine, using aprobarbital as internal standard. Whole‑blood samples are used for the analysis and solid‑phase extraction is applied for sample pretreatment. Samples of 10 µl are injected onto a 4 µm Novapak C18 column (3.9×300 mm) with a column temperature of 40 °C. The drugs are eluted with a mobile phase containing potassium phosphate buffer (pH 7.0), acetonitrile and methanol (110:50:30, v/v/v) at a flow rate of 0.5 ml/min. Signals are monitored by a photodiode‑array detector at 210 nm with a bandwidth of 10 nm. These three antiepileptic drugs and their principal metabolites are well separated within 15–20 minutes. Within‑day coefficients of variation are between 9.4 and 13.8 and between‑day coefficients between 4.11 and 11.94. Recovery ranges from 60 to 125 %. The method allows evaluation of forensic blood samples for detection and quantitation of these drugs.
